p53 mutants are found in more than half of human cancers and are considered as the most important human cancer related gene. p53 is detected at 53kD position by electrophoresis and is composed of 393 amino acids. In the unstressed normal cells the p53 level is low and it is inactive. However with stress especially with DNA damage it is activated to promote arrest of cell cycle and repair of DNA damage or induction of apoptosis. The functions of p53 are regulated by phosphorylation of serine and threonine and acetylation of lysine at various sites in the molecule. Among the phosphorylation sites Ser46 is phosphorylated when DNA damage is so severe as to become unrepairable which leads to apoptosis by activating transcription of proapoptotic genes such as p53AIPI As to the kinase of phosphorylation of Ser46 involvement of DYRK2 and ATM have been implicated.

Ser315 is phosphorylated by aurora kinase and cycline-dependent kinases when cells are subjected to stress such as DNA damage and microtubule disruption by nocodazole (ref 1 2 & 3). However the effect of the phosphorylation on the function of p53 is mostly unknown.
This product is the purified IgG fraction obtained from serum-free culture medium of mouse hybridoma (clone #18) which produces monoclonal antibody that specifically recognizes human p53 protein with phospholyrated Ser315.

MdmX (synonyms: Mdm4 HdmX) inhibits p53-and p73-dependent cell cycle arrest and apoptosis by binding to the transcription activation domains of these proteins. MdmX consists of 490 amino acids with the molecular weight of 54864 and contains a RIING-finger domain and a nuclear transport signal. It is known that the protein migrates aberrantly in SDS-PAGE at the position of an 80-kDa protein. MdmX is phosphorylated at Ser367 by Chk2 kinase downstream of ATM in response to DNA damage and as a result it binds to14-3-3 and is transported into nucleus where it is degraded by Mdm2. This process activates the p53 functions (1 2 and 3).

E2F1 is a member of E2F group of proteins that share common structural and functional domains and plays a major role during the G1/S transition in the mammalian cell cycle as a transcriptional factor (1). E2F1 is regulated during cell cycle progression. It is phosphorylated at Ser364 by Chk2 kinase in response to DNA damage stabilized mobilized to nucleus and activated as a transcription factor (2). It induces apoptosis by activating transcription of the p53 homolog p73 (3). E2F1 protein consists of 437 amino acids with a molecular mass of 46 kDa.
