Backgrounds:DNA polymeraseβ is a distributive polymerase involved in base excision repair which repairs damaged DNA by excising modified bases (oxidized methylated deaminated etc.) (ref. 1).This product is highly purified full-length rat DNA polymeraseβ overproduced in E. coli with high enzymatic activity without any tag attached (ref.2).

Applications
1) For the studies on the mechanisms of base-excision repair of DNA damage
2) As a positive control of western blotting with anti-DNA polymeraseβ antibody

Properties of the product
Enzyme activity: 90 unit/ul (1unit of the enzyme activity incorporates 1 nanomole of dNTP into acid-insoluble fraction at 37℃ in 60 min.)

DNA polymerase delta is one of the three eukaryotic DNA polymerases which are essential for chromosome replication and is also involved in nucleotide excision repair base excision repair and VDJ recombination (1 2). Subunit 3/p66 is a functionally important subunit of human polymerase delta which stabilizes polymerase delta complex and increases the affinity of polymerase delta for PCNA (3).
This product is the IgG fraction purified from serum-free culture medium of mouse hybridoma (2A1C11) by propriety chromatography under mild conditions.

PCNA (Proliferating cell nuclear antigen) is a homotrimeric protein (261 aa; 29 kDa) known to act as a co-factor for DNA polymerase  which is responsible for leading strand DNA replication. PCNA was originally identified as an antigen that is expressed in the nuclei of cells during the DNA synthesis phase of the cell cycle. Crystal structure data suggests that a PCNA homotrimer ring encircles and slides along the DNA double helix. Multiple proteins involved in DNA replication DNA repair and cell cycle control bind to PCNA rather than directly associates with DNA thus facilitating rapid processing of DNA. PCNA is a useful marker for DNA synthesis and some cancers. It is highly conserved among most amimals.
Applications confirmed:
1. Functional studies on DNA replication recombination and repair. (Ref 2 3 5 6 7 8 9 10).
2. Identification of proteins interacting with PCNA by using PCNA –conjugated resin. (Ref 1 5)
3. Ubiquitination targets (Ref 4 9 10).
4 SDS-PAGE (Fig. 1). 5. Western blot (Fig. 2) . 6. Dot blot. 7. ELISA. Not tested for other applications.

Function: Ribonucleoside-diphosphate reductase subunit M2 (RRM2; 389 aa
45 kDa) also known as ribonucleotide reductase subunit R2 (RNR-R2) is a
rate-limiting subunit of an enzyme that catalyzes the formation of
deoxyribonucleotides from ribonucleotides. Deoxyribonucleotides in turn
are used in the synthesis of DNA.

Immunogen: Synthetic peptide (11 aa) in the C-terminal region of Human
and Mouse RRM2.

Application: WB IP IF IHC
