Resume of DNA synthesis which was halted by various physical and chemical factors was triggered by repair system including Rad6. Rad6 (152a.a 17.3kDa) is the ubiqutin-conjugated enzyme E2A and complex with two Rad18 molecules ubiquitin ligase E3 to ubiquitinate PCNA by utilizing poly-ubiquitin chain that HLTF-UBC13 synthesized in translesion DNA synthesis (TLS) and template switch (TS). Two isoforms of RadA Rad6A and Rad6B differs in only 6 amino acids. It also plays an important role in epigenetic transcriptional regulation by catalyzing the monoubiquitination of histone H2B at Lys-120 for a formation of H2B K120 ub1.

Unlike ubiquitination which targets proteins for degradation SUMO modification plays a critical role in a number of cellular functions including nucleocytoplasmic transport gene expression cell cycle and formation of subnuclear structures such as promyelocytic leukemia (PML) bodies. There are three confirmed SUMO isoforms in human; SUMO1 SUMO2 and SUMO3. SUMO2 /3 show a high degree of similarity to each other and are distinct from SUMO1. Individual SUMO family members are all targeted to different proteins with diverse biological functions. SUMO1 is encoded as a 101 aa protein and first Met and C-terminal 4 aa are removed from the preprotein.

Function: DNA polymerase eta ( 713 aa 78 kDa) is specifically involved in DNA repair. Plays an important
role in translesion synthesis where the normal high fidelity DNA polymerases cannot proceed and DNA
synthesis stalls. Plays an important role in the repair of UV-induced pyrimidine dimers. Depending on the
context it inserts the correct base but causes frequent base transitions and transversions. May play a role
in hypermutation at immunoglobulin genes. Forms a Schiff base with 5-deoxyribose phosphate at abasic
sites but does not have lyase activity. Targets POLI (Pol iota) to replication foci.
Involvement in disease: Xeroderma pigmentosum variant type (XPV) [MIM:278750]: An autosomal
recessive pigmentary skin disorder characterized by solar hypersensitivity of the skin high predisposition
for developing cancers on areas exposed to sunlight and in some cases neurological abnormalities. XPV
shows normal nucleotide excision repair but an exaggerated delay in recovery of replicative DNA
synthesis. Most patients with the variant type of xeroderma pigmentosum do not develop clinical
symptoms and skin neoplasias until a later age. Clinical manifestations are limited to photo-induced
deterioration of the skin and eyes.
