Background: Heparin-binding epidermal growth factor-like growth factor (HB-EGF) is synthesized as a membrane-anchored precursor that is proteolytically cleaved to release the soluble mature growth factor HB-EGF (1 2). The former functions as juxtacrine and the latter as paracrine growth factor. HB-EGF activates EGFR and ErbB4 and promotes the development of many tissues. In human ProHB-EGF is the cellular receptor for diphtheria toxin (3). Non-toxic mutant of diphtheria toxin CRM197 inhibit HB-EGF function which is elevated in most ovarian cancer is being tested as an anticancer drug (4).

Anti-SLA (haploid sperm cell-specific antigen) antibody clone TRA54

Many transcripts for proteins specifically expressed in haploid germ cells are synthesized in the spermatogenesis. A monoclonal antibody TRA54 recognizes specific organelles in germ cell cytoplasm from spermatocytes to spermatids (1 2). Western blots using TRA54 revealed broad bands with 85 kD 190 kD and >200 kDa in testis probably due to alternative splicing and/or post-translational modification (Fig. 1). The molecular size diversity is due to the difference in glycosilation (Ref. 1). This antibody can be used for immunohistochemical staining of testicular tissues (1 3).
The hybridoma producing TRA54 was obtained from the immunaization of a rat with cell lysates of adult mouse testes by the group of Prof. Yoshitake Nishimune of Osaka University.

Mutations in the octapeptide repeat regions as well as elsewhere in this gene have been associated with neurodegenerative diseases such as Creutzfeldt Jakob disease fatal familial insomnia Gerstmann Straussler disease Huntington disease like 1 and kuru. The infectious isoform of PrPC known as PrPSc is able to convert normal PrPC proteins into the infectious isoform which is insoluble amyloid aggregate by changing their conformation (1).
Mature PrP protein in human consists of 209 amino acids. Several topological forms exist; one cell surface form anchored via glycolipid and two transmembrane forms therefore multiple bands are observed in SDS-PAGE (Figure).
