Tem1 is a low-molecular-weight GTP-binding protein (GTPase) which is required for the termination of M phase in cell division. The defect of Tem1 was lethal and the Tem1-defective cells were arrested at telophase with high H1-kinase activity indicating that Tem1 is required to exit from M phase. The defect of Tem1 was suppressed by a high dose of cdc15 which encodes a protein kinase. Tem1 functions upstream of cdc15 kinase and may be required to activate the cdc15 protein kinase pathway. A cascade consisting of　Tem1 and kinases act to terminate mitosis.

Heterogeneous nuclear ribonucleoprotein U (hnRNP-U also known as scaffold attachment factor A SAF-A) is a nuclear matrix-associated protein that interacts with chromosomal DNA. hnRNP-U specifically binds to scaffold/matrix attachment region of DNA and could thus be involved in higher order chromatin structure. hnRNP-U is also a RNA binding protein and forms complexes with heterogeneous nuclear RNA (hnRNA) and plays an important role in pre-mRNA processing and transport.
HnRNP-U is reported to interact with necdin a growth suppressor that is expressed in terminally differentiated neurons and skeletal muscle cells. It has been shown that hnRNP-U recruits necdin to the nuclear matrix where they form a stable complex. It is suggested that necdin suppresses cell proliferation through its interaction with hnRNP-U in the specific subnuclear structure (ref.2).

Mitf (Microphthalmia-associated transcription factor) is a transcription factor that contains both basic helix-loop-helix and leucine zipper structural features. It plays a critical role in the differentiation of various cell types such as neural crest-derived melanocytes mast cells osteoclasts and optic cup-derived retinal pigment epithelium. Mutations in Mitf cause auditory-pigmentary syndromes such as Waardenburg syndrome type 2 and Tietz syndrome. Alternatively spliced transcript variants encoding different isoforms have been identified.
The antibody was produced by immunizing rabbit with recombinant human Mitf protein in the laboratory of Prof. H. Yamamoto.

Both necdin and nucleobindin 2 are expressed in differentiated neurons and skeketal muscles and these proteins are likely to be involved in the regulation of survival and death of postmitotic cells by controlling Ca2+ homeostasis

Anti-Nuf2 antibody rabbit serum

Nuf2 is a component of kinetochore-associated Ndc80 complex which serves to attach microtubules to the kinetochore. Nuf2 is a conserved protein from yeast to human. Disruption of the Nuf2 gene in Schizosaccharomyces pombe causes defect in chromosome segregation and in the spindle checkpoint. Yeast Nuf2 disappears from the centromere during meiotic prophase when centromeres lose their connection to the spindle pole body and plays a regulatory role in chromosome segregation. In human cells Nuf2 specifically functions at kinetochores for stable microtubule attachment. Down regulation of the protein by RNA interference results in failure of the kinetochores to form attachments to the spindle microtubules. As a result cells are blocked in the prometaphase stage with an active spindle checkpoint and undergo cell death.

Background: Proliferating cell nuclear antigen (PCNA) is a 29 kDa homotrimeric protein known to act as a co-factor for DNA polymerase delta which is responsible for leading strand DNA replication. PCNA was originally identified as an antigen that is expressed in the nuclei of cells during the DNA synthesis phase of the cell cycle. Multiple proteins involved in DNA replication DNA repair and cell cycle control bind to PCNA rather than directly associating with DNA thus facilitating fast processing of DNA.

DDX18 protein (670aa 75.4KDa) is a putative ATP-dependent RNA helicase since it possesses a DEAD box motif characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD).RNA helicases which may play a role in ribosome biogenesis. DDX18 acts as a cofactor for XPO1-mediated nuclear export of incompletely spliced HIV-1 Rev RNAs.
